Each freeze-thaw cycle stresses the peptide — ice crystal formation, and the concentration effects at the interface as water freezes around the solute, both contribute to aggregation and to adsorption losses onto container surfaces. A single freeze and one thaw for immediate use is different from repeatedly freezing and thawing the same aliquot.
04A practical framework
If a large vial has been reconstituted, the standard advice is to freeze it in single-use aliquots immediately, rather than freezing and re-freezing one working vial. That converts "many freeze-thaw cycles on one sample" into "one freeze-thaw cycle each on many smaller samples" — the same total material, but a much smaller degradation cost per unit used.
